What Pure Peptides UK's COAs, HPLC and mass spec reports actually show, whether batch data is lot-specific, and how £15–£60 vial pricing compares.

Pure Peptides UK is a UK-based supplier of research-use-only peptide reference materials, and a meaningful review of it rests on four documents rather than four stars: the certificate of analysis, the HPLC chromatogram behind that COA, the mass spectrometry report confirming identity, and the lot record tying both to the vial that arrives. Suppliers in this segment — Pure Peptides UK among them — typically advertise per-batch purity of ≥98% by HPLC, ship lyophilized material at ambient temperature, and list single vials somewhere in the £15–£60 band. Whether that is adequate for a given laboratory depends on three things the marketing page rarely answers: whether the COA is batch-specific rather than representative, whether an independent lab generated it, and whether net peptide content is stated at all. Usually it is not.

What a supplier review actually measures

Star ratings on a research-chemical vendor measure order fulfilment. They tell you the parcel arrived, the vial was intact, and the customer service email got answered. None of that is analytical evidence. A reviewer who received 47 vials without incident has demonstrated logistics, not identity.

The variables that determine whether a reference material is usable in a documented experiment are narrower and duller:

  • Identity — does mass spectrometry return the expected monoisotopic or average mass, typically within ±0.5 Da on a standard ESI-MS instrument, or under 10 ppm on a high-resolution system?
  • Purity — what percentage of the integrated peak area at 214 nm (the peptide bond absorbance) belongs to the target peak, and what are the named impurities?
  • Net content — of the stated 5 mg or 10 mg in the vial, how many milligrams are actual peptide?
  • Traceability — does the lot number on the vial appear verbatim on the COA, and can the supplier reproduce that COA on request 18 months later?
  • Stability — what storage temperature and shelf life is the material qualified for, and is that claim supported by data or asserted?

A supplier that scores well on the first four and poorly on shipping speed is a better analytical partner than the reverse.

Reading the certificate of analysis properly

Most COAs circulating in this market are PDFs with a company logo, a compound name, a percentage, and a signature block. Several failure patterns show up repeatedly.

Representative versus batch-specific. A "representative" COA describes a batch that is not the batch you received. It is a specimen document. If the lot number on the certificate does not match the lot printed on the vial label, the certificate documents nothing about your material. Ask directly whether COAs are regenerated per lot.

Chromatogram or nothing. A purity figure without the underlying trace is unverifiable. A real chromatogram shows the retention time, the gradient, the column chemistry (usually C18, 5 µm particle, 4.6 × 250 mm), the injection volume, and — critically — the baseline. Broad unresolved shoulders next to the main peak can be integrated generously. Seeing the trace lets an analyst judge that themselves.

Who ran it. In-house testing is not disqualifying, but it is a different evidentiary class than a third-party report. Independent HPLC/MS on a single sample runs roughly $150–$400 per compound at a contract lab in the US, with 5–10 business day turnaround. That is the price of converting a supplier claim into your own data.

A purity percentage with no chromatogram, no lot number, and no named laboratory is a marketing figure that happens to have a decimal point in it.

Net peptide content: the number almost nobody publishes

This is the single most consequential gap in how research peptides are sold, and it rarely appears in supplier reviews.

Synthetic peptides purified by reverse-phase HPLC are typically lyophilized as salts. The counterion — most often trifluoroacetate (TFA) from the mobile phase, sometimes acetate after a salt exchange — remains bound to basic residues. Residual water is also present. The result is that the gross mass in the vial is not peptide mass.

Realistic composition of a "10 mg" lyophilized vial:

  • Peptide: often 75–92% of gross mass, depending on the number of basic residues
  • Counterion (TFA or acetate): commonly 5–15% of gross mass
  • Water, by Karl Fischer titration: commonly 3–8%

A vial labelled 10 mg at 99% HPLC purity can therefore contain roughly 8.0–8.5 mg of actual peptide. The 99% figure is still accurate — it describes chromatographic purity of the peptide fraction, not the fraction of the powder that is peptide. Two different questions, routinely conflated.

The instrument that answers the second question is amino acid analysis (AAA), or quantitative NMR. Very few suppliers in the UK or US research-materials segment publish either. Asking for AAA data is one of the fastest ways to distinguish a supplier with real analytical infrastructure from one reselling a distributor's stock with a rebranded PDF. Traceability, storage, and stability

Ambient-temperature shipping of lyophilized peptide is standard practice and generally defensible — dry solid peptide tolerates several days at room temperature far better than the same material in solution. The claims worth scrutinising concern what happens after delivery.

Commonly cited storage conditions for lyophilized material:

  • –20 °C: standard long-term storage, with stated shelf life frequently 24–36 months
  • –80 °C: used for extended archival storage of sensitive sequences
  • 2–8 °C: shorter-term, typically quoted at weeks to a few months
  • Ambient: transit only, usually qualified for 7–14 days

The question to put to any supplier: is the 24-month figure backed by stability data on that compound, or is it a category default applied to the whole catalogue? Both answers are common. Only one is evidence. A supplier that says plainly "this is a catalogue-wide default, we have not run accelerated stability on this sequence" is more trustworthy than one that implies otherwise.

Traceability has a simple test. Email the supplier a lot number from an order placed a year ago and ask for the COA. A vendor with genuine batch records returns it in a day or two. A vendor without them sends a current representative document, or nothing.

Cost: what you actually pay per usable milligram

Headline vial prices mislead in both directions. The realistic landed cost for a US laboratory buying from a UK supplier includes several line items:

  • VAT at 20% — generally removable on export to the US, but only if the supplier zero-rates correctly at checkout; some do not, and reclaiming it afterwards is slow
  • International shipping — commonly $25–$60 per parcel, with 5–12 business day typical transit
  • Customs handling and duty — the $800 Section 321 de minimis exemption that US importers long relied on was eliminated in 2025, so low-value parcels can now attract duty and broker fees that previously did not apply. Confirm current treatment before assuming a small order clears free
  • Independent verification — the $150–$400 per compound noted above, if you intend to trust the material
  • Failed-lot risk — a rejected batch costs the vial price plus the verification cost plus the calendar time

When this supplier — or any offshore supplier — is not worth it

This is the part most reviews omit, so it is worth being blunt.

Do not buy from an overseas research-materials supplier if any of the following apply:

  • Your work requires GMP or pharmacopoeial-grade material. Research-grade peptide is not GMP. No amount of HPLC purity makes it so. If your protocol, funder, or institution requires GMP documentation, this entire market segment is the wrong place to shop, and buying here wastes the budget line.
  • You need cGMP-comparable regulatory filings. No RUO supplier can support an IND or equivalent submission. The research use only designation is a hard boundary, not a formality.
  • Your timeline is under two weeks. International transit plus customs realistically consumes 7–21 days, and delays of 30+ days on customs holds are not rare. Domestic sourcing costs more and arrives sooner.
  • You cannot afford independent verification. If the budget does not accommodate $150–$400 per compound for third-party confirmation, you are relying entirely on a supplier-generated PDF. That may be an acceptable risk for exploratory work and is a poor one for anything you intend to publish.
  • You need a certified reference material with an assigned uncertainty value. RUO peptide is not a CRM. Metrologically traceable standards come from a small number of accredited producers and cost 5–50× more, for reasons that are real.
  • You require net peptide content and the supplier won't provide AAA data. For quantitative work, gross mass is not a usable input. Missing AAA is a legitimate reason to walk away.

Additional limitations that apply even when the purchase makes sense: batch-to-batch variation is normal in solid-phase synthesis, so a good result on lot A does not guarantee lot B; fill weights on small vials commonly vary by ±5%; and endotoxin is usually untested unless explicitly requested, with <0.5 EU/mg by LAL being the figure to ask for when it matters. Returns on opened vials are generally not accepted anywhere in this market — read the returns terms before ordering, not after.

Qualifying the supplier in a single order

A pragmatic first purchase treats the order as a supplier audit rather than a stock replenishment.

  • Order one vial of a single compound, not a catalogue spread
  • Request the batch-specific COA before payment, and check the lot number against the vial on arrival
  • Ask for the raw chromatogram, not the summary PDF
  • Ask, explicitly, for net peptide content and whether AAA was performed
  • Send the vial for independent HPLC/MS and compare the impurity profile, not just the headline percentage
  • Twelve months later, email the lot number back and see whether the record still exists

Six data points, one vial, roughly $200–$500 all-in. That produces a defensible answer about Pure Peptides UK, or about any supplier, that no aggregate star rating can.
